1. 洗滌緩衝(chong) 液的製備與(yu) 儲(chu) 存
手冊(ce) 中明確要求HDL重懸緩衝(chong) 液需新鮮製備且不可儲(chu) 存。對於(yu) 1×HDL洗滌液，我個(ge) 人建議每次新鮮配製，但將其置於(yu) 4°C儲(chu) 存直至使用是可行的，無需過度擔憂。
2. PBS配方建議
在此步驟中，您可根據下遊應用需求選擇適合的緩衝(chong) 液進行透析。目前未提及具體(ti) PBS配方（如pH、離子強度、是否含Ca²⁺/Mg²⁺或防腐劑如疊氮鈉）的強製要求，但需確保其與(yu) 後續實驗兼容。
3. 透析膜選擇
對於(yu) 貨號Cat# STA-607，透析方案可采用任意透析方法。例如，可使用賽默飛世爾的Slide-A-Lyzer MINI透析裝置。我們(men) 通常推薦使用10 kDa截留分子量（MWCO）的透析膜，但其他尺寸也可接受。操作流程需遵循說明書(shu) ，一般步驟如下：
• 用300倍樣本體(ti) 積的緩衝(chong) 液透析2-4小時；

5. 透析後樣本情況
• 最終體(ti) 積：Cat# STA-607的典型產(chan) 量約為(wei) 7 ml（來自人血清或血漿樣本）；
• 蛋白濃度範圍：純化後HDL的濃度約為(wei) 4500 µg/ml。
6. 透析後樣本處理
需注意，Cat# STA-607提供的溶液並非無菌，因此我們(men) 從(cong) 未對純化後的HDL進行過過濾除菌。理想情況下，純化後的脂蛋白可長期儲(chu) 存於(yu) 4°C；若樣本含50%甘油，則可儲(chu) 存於(yu) -20°C。關(guan) 於(yu) 分裝體(ti) 積，我們(men) 無法給出具體(ti) 建議，需客戶根據下遊應用自行決(jue) 定。
7. 蛋白濃度測定方法推薦
手冊(ce) 未明確蛋白濃度測定方法，建議客戶根據下遊應用選擇合適方法。我們(men) 通常采用BCA assay測定總蛋白濃度。
8. 總蛋白濃度與(yu) HDL濃度的等效性
遺憾的是，我們(men) 從(cong) 未將BCA assay測定的濃度與(yu) ELISA結果進行過比較（僅(jin) 通過ELISA確認ApoB的存在）。因此，無法提供校正方法或參考標準。關(guan) 於(yu) 殘留的非HDL蛋白，我們(men) 通過SDS-PAGE評估純度。
9. ApoM穩定性
很遺憾，我們(men) 從(cong) 未使用Cat# STA-607評估過ApoM的穩定性（包括其在硫酸葡聚糖沉澱、離心、重懸、洗滌及透析過程中的結合情況），也無相關(guan) 數據證明其未解離。
10. S1P穩定性
同樣，我們(men) 從(cong) 未評估過ApoM或S1P在純化過程中的穩定性（如離心、渦旋/重懸、透析等步驟是否會(hui) 導致S1P與(yu) ApoM解離或部分丟(diu) 失）。
11. HDL顆粒完整性
該試劑盒旨在維持HDL顆粒的天然結構（包括脂質雙層及載脂蛋白組成），但我們(men) 從(cong) 未對其結構或功能進行過驗證（如電子顯微鏡、天然凝膠電泳等數據）。
12. ApoM-S1P-HDL的沉澱行為(wei)
我們(men) 從(cong) 未評估過ApoM或S1P與(yu) HDL的結合情況，因此無法確定ApoM-S1P-HDL是否會(hui) 與(yu) 總HDL共同沉澱，或是否存在選擇性丟(diu) 失的風險。
13. 最終產(chan) 物的組成
按照手冊(ce) 操作獲得的最終產(chan) 物是否為(wei) “總HDL"？是否包含攜帶ApoM的HDL亞(ya) 群（即ApoM陽性HDL）？若有，是否有參考數據（如文獻或內(nei) 部驗證）說明其在總HDL中的比例？
很遺憾，我們(men) 從(cong) 未評估過ApoM或S1P的相關(guan) 情況，無法回答此問題。此外，關(guan) 於(yu) 純化產(chan) 物中是否殘留其他脂蛋白（如LDL、VLDL）或血漿蛋白，我們(men) 通過SDS-PAGE評估純度。
14. 直接兼容性
純化後的HDL產(chan) 物（透析後）是否可直接用於(yu) ApoM免疫沉澱？是否需要額外的緩衝(chong) 液交換或純化步驟（如脫鹽、超濾）？
很遺憾，我們(men) 從(cong) 未評估過ApoM或S1P的相關(guan) 情況，無法給出建議。
15. 殘留幹擾成分
純化後的HDL產(chan) 物中是否有殘留成分（如硫酸葡聚糖、Tris、沉澱試劑等）可能幹擾抗體(ti) -抗原結合？這些殘留是否會(hui) 影響免疫沉澱效率？
通過使用硫酸葡聚糖去除液並進行透析，可去除大部分幹擾成分。
