Q:Dear Technical Support Team, I am reaching out for assistance regarding the use of your DirectPCR Lysis Reagent (Mouse Tail, Cat#102-T) in our mouse genotyping experiments. We use mouse toe samples instead of mouse tails as the starting material for lysis with this reagent. Previously when we used this product, the toe samples remained fully intact after the lysis procedure(60℃ 16h; 85℃ 45min), and we consistently obtained high-quality, reliable genotyping results. Two years ago, we suddenly failed to obtain any target genotyping bands, which was most likely caused by the expiration of that specific bottle of your reagent. We purchased a new bottle of DirectPCR Lysis Reagent (Mouse Tail, Cat#102-T) last November. Now we are able to get bands with this new batch, but the bands are abnormally larger than expected. We also noticed that the toe samples are no longer intact after lysis: all the soft tissues are mostly digested, with the toe bones remaining visible after the lysis step. Occasionally, the crude lysate appears viscous. We would like to consult you on the following questions: 1. Are these observations (bone visible and crude lysate appears viscous) normal phenomenon when using this reagent for toe samples? Or does it indicate that our samples are over-digested? 2. If this is over-digestion, could it be the cause of the abnormally large molecular weight of the bands we observed? We would highly appreciate your guidance on these issues. A:Dear Team,
Thank you for your detailed description of the issue. Based on what you reported, the digestion pattern you are seeing with toe samples is generally consistent with normal DirectPCR performance. When toe tissues are lysed with Proteinase K, it is expected that most soft tissues will be digested efficiently while harder structures (e.g., bone) remain visible. Likewise, a viscous crude lysate is commonly observed when lysis is successful, as released genomic can increase viscosity. These findings do not, by themselves, indicate “over-digestion," and extended incubation (such as 16 hours) typically does not compromise routine genotyping because moderate fragmentation does not materially affect amplification of standard PCR-sized amplicons.
Regarding the abnormally larger-than-expected bands, over-digestion is very unlikely to be the cause, since the lysis step does not create PCR products that migrate at a higher apparent molecular weight. A band size shift is more commonly explained by PCR-related factors such as non-specific amplification, primer design or primer mix-ups, altered cycling conditions, polymerase choice, gel percentage/ladder selection, or the presence of inhibitors that can affect amplification behavior. As a first check, we recommend confirming the expected amplicon size from the primer design, running a purified genomic control with the same primer set, and verifying your ladder and gel conditions.
For best reproducibility with DirectPCR lysates, please also ensure that you are using a hot-start 韦德唯一官方 polymerase and that lysate input is kept low (typically 0.5-1.0 µl lysate per 50 µl PCR reaction; excess lysate can introduce inhibitors without improving yield). In addition, if your tubes are incubated without mixing, increasing reagent volume and/or providing rotation/shaking during lysis can improve consistency across samples, as incomplete or heterogeneous lysis can sometimes lead to variable PCR behavior.

I understand that hemoglobin and hemin are different molecules. Could you please confirm whether your kit is designed or validated to detect hemin specifically?
Sample type for detection: Regarding the detection of heme (or hemin) in cells, does your kit require the use of cell lysates after hemin treatment? Or are other sample types (such as culture medium) also applicable?

My apologies for any confusion - in your original mail you mentioned the customer wanted to test the kit with hemoglobin, so that is what we looked into. However, if the customer is actually interested in measuring Hemin, we will need to look into that as well.
